Is it possible to get a report of traffic between 2 specific interfaces?

exactly what are you looking for?

are you looking for utilization between the two interfaces, or what application are traversing between the two?

And what type of interfaces? Are these router interfaces or server interfaces? What information do you want in the report? More info is needed to answer that question.

An interface on a switch to an interface on another switch.

Basically there is a link between 2 sites and they want to see how much traffic is going across it.

You should be able to get this information by just monitoring one of the switch interfaces via SNMP. If you monitor both interfaces, you could compare the numbers and they should be mirror images of each other.
