Is there a way to Poll Registry Keys or look for a value in a text file?

You can do that in SAM today with custom script monitors or there is also a relatively new module for Orion, Server Configuration Monitor, that is designed almost precisely for that use case: Server Configuration Change Monitoring Tool | SolarWinds

I'd like to be able to monitor if a registry key is present or its value changes.

We do have application templates for monitoring log files on Windows and *nix hosts. These are included as part of SAM 5.0, or can be download from the Thwack Content Exchange.

We don't currently have built-in method for monitoring registry keys but if you could provide me with your use case I'll be happy to log this as a feature request. The only way to do this today would be though the use of one of our script monitors.
