Do the users at this other domain live on the same Exchange server?
Please check your Exchange receive connectors (EMC > Server Configuration > Hub Transport > Receive Connectors). Ensure that you have an entry for the WHD server with the WHD server's IP address listed on the Network tab (example below--192.168.13.20). Make sure that Anonymous users is checked on the Permission Groups tab.
Additionally, if you are trying to relay to an external server, please check the Send Connectors for that domain (EMC > Organization Configuration > Hub Transport > Send Connectors tab). When creating/editing the Send Connector, be sure to check the Network tab so that mail will be appropriately routed.

I created a record under accepted domains and configured it as an external relay, this seems to have resolved my issue here.
I know this is a REALLY old post but wondering if someone can help me with the same issue.
I followed the guide as above but we are running Exchange 2010. I am running the latest version of the WHD as well 12.5.1. But still unable to relay.
if it is the same type of error message (e.g. 550 5.7.1) then you'll need to configure your Exchange environment to allow SMTP relay. You can allow anonymous access to that relay or not; if not you'd require authentication to be able to relay.

I have added in the IP addy to allow relay as above, but still seem to get the same error . I rebooted the WHD as well to see if this helped but still no .
I found this link and followed this. It resolved my issue completely. It does say its for 2007 but I done this with Exchange 2010 and they were the EXACT Same steps.
